WP2: CMEs and Workshops

To gain knowledge in renal disease in patient with obesity, metabolic syndrome and diabetes, the Coordinator and partners will work together in the organization of CMEs and workshops. We will collaborate with the Diabesity working group of the ERA, a group whose aim is to disseminate knowledge on this field (https://www.era-online.org/en/diabesity/). With these activities we will promote networking, interchanges of ideas, design common projects, publications, etc. We expect an audience of 60-80 assistants in each CME and around 20 assistants per workshop.

We will organize 3 CMEs in Tenerife: one per year on areas related to the topic of the proposal

  • CME 1: Gender Differences in Renal Disease in Diabetes and Obesity
  • CME 2: Renal Histology in patients with diabetes, obesity and metabolic síndrome
  • CME 3: Exercise and dietary interventions in patients with renal disease, diabetes and obesity

and four workshops:

  1. Animal models of Renal Disease in Obesity and Diabetes
  2. Application of the poulation PK in the clearance of iohexol
  3. Renal lipotoxicity, from cells, to animal models and humans
  4. Research proposal for EU grants 

WP3: Visibility

  • Workshop I: Measured GFR by iohexol DBS in humans
  • Workshop II: Measured GFR by iohexol-DBS in animal models
  • International consensus on obesity and MS in renal transplantation

WP4: Stafff exchange – Expert visits

We will establish interchanges between scientists and staff of the coordinator and partners to enhance, reinforce and boost the scientific skills of our group. This will also help discussing collaborative projects. All these will promote our participation in international grants i.e. HORIZON EUROPE. Finally, this is crucial to create long standing collaborations that persist beyond the project. We plan 6 expert visits aimed at lecturing and teaching in specific topics. We plan 3 staff interchanges per year in which members of our group (1-2 persons) will go to the labs of the partners from 1 to 2 weeks (depending on the topic).

The Technical Experts visits are:

  1. Epidemiological/population based studies in Nephrology
  2. Clinical Trials in Post-Transplant Diabetes Mellitus
  3. Population Kinetics applied to the clearance of iohexol
  4. Academia-Industry joint projects in nephrology
  5. Chronic liver disease, NASH, and renal damgare
  6. New drugs in diabetic nephropathy

Planned staff interchanges are:

  • Improving lipidomic analysis (Copenhagen)
  • Population PK of drugs (Oslo)
  • Techniques in animal models of diabetes/obesity (Copenhagen)
  • Clamp techniques to evaluate insulin resistnace-sensitivity(Oslo/Copenhagen)
  • Exercise studies in renal and non renal disease (Copenhagen)

WP5: Research project – Renal disease in the continuum of obesity-metabolic syndrome and diabetes

We will analyze diverse aspects of renal disease in obesity, metabolic syndrome and T2DM, in particular:

A.- RENAL MORPHOLOGY: We will collaborate with the European Nephrectomy Bio-Bank (ENBiBA) project. It studies patients who will undergo a nephrectomy (mainly due to cancer), characterizes the phenotype i.e. normal, overweight, obese, metabolic syndrome, prediabetes and diabetes; collects clinical variables, serum and urine samples before nephrectomy and obtains after the nephrectomy a large piece of renal tissue of the unaffected area, distant from the tumor. The cases will be grouped in normal (lean), overweight, obese, metabolic syndrome, diabetes with or without proteinuria. We will evaluate gender differences in renal histology and pathogenic factors in the studied conditions.
B.- RENAL LIPOTOXICITY: Inmuno-histochemistry: using adipophilin stain, to evaluate lipid deposits. Lipidomics: in fresh tissue, the presence of fatty acids by mass spectrometry (a set of 30 acids), lipid classes (sphingolipids, sphingomyelin, phosphatidylcholine, ceramides, etc) and fatty acids in the lipid classes. Also, we will set up, in collaboration with CPN the technique of shotgun lipidomics. Urinary lipidomics: based on the finding of the lipidomics in fresh tissue we will look for lipids in the urine by mass spectrometry.
C.- INFLAMMATION IN RENAL TISSUE AND URINE: we will determine inflammatory markers (TNF-a, IL-6, MCP-1, VCAM-1 and NFkB-p65) in renal tissue and in urine. In particular we will look for links between lipid deposits and a pattern of inflammatory deposits in the different groups analyzed.
